Aston Martin unveiled the V12 Vanquish at the Geneva Motor Show in March 2001. Ian Callum's styling set the template for later Aston Martins. The 5.9-liter V12 made 460 bhp in its earliest form. The Vanquish was the first Aston Martin with an automated manual transmission and paddles. The Vanquish S arrived in 2004 with 520 bhp. It was the last model largely hand-built in the old Newport Pagnell factory. Production ran from 2001 through 2007, with approximately 2,578 examples built.

The second-generation Vanquish was introduced in 2012 on the VH platform. Aston Martin built it in Gaydon to replace the discontinued DBS. It had an aluminum structure, an all-carbon-fiber body and a 6.0-liter V12. Output was 565 bhp. A Vanquish S with a more powerful engine was launched in 2017. The DBS Superleggera succeeded the Gaydon Vanquish in 2018.

In September 2024, Aston Martin announced the third-generation Vanquish. The 2025 model has a 5.2-liter twin-turbo V12 rated at 835 PS. Power goes to the rear wheels through a ZF eight-speed automatic. The V12 has the highest specific power output of any powertrain in Aston Martin's 100-plus-year history. MotorTrend says the 2025 Vanquish replaces the DBS 770 Ultimate.
